蛋白构建

A DNA sequence encoding the Human IL2RB (P14784)(Ala27-Asp239) was fused with a flag tag at the C-terminus, constructed the plasmid 1; A DNA sequence encoding the Human IL2RG (P31785) (Leu23-Asn254) was expressed with a polyhistidine tag at the C-terminus, constructed the plasmid 2. The two plasmids were co-expressed and the IL2RB & IL2RG heterodimer was purified.

The recombinant heterodimer of human IL2RB & IL2RG comprises 616 (313+303) amino acids and has a calculated molecular mass of 68.34 (34.25+34.09) KDa. The apparent molecular mass of human IL2RB & IL2RG heterodimer is approximately 59 kDa and 45 kDa respectively in SDS-PAGE. The molecular weight of this protein is around 98.7 kDa verified by SEC-MALS(Routinely tested).
